(Rac)-LM11A-31 is a p75 NTR modulator for diabetic retinopathy research

**Background**

The p75 neurotrophin receptor (p75 NTR) is a member of the tumor necrosis factor receptor superfamily and plays a critical role in regulating neuronal survival, apoptosis, and axonal growth. In the context of metabolic diseases, the dysregulation of p75 NTR signaling is closely linked to the development of diabetic complications. Specifically, the binding of pro-nerve growth factor (proNGF) to p75 NTR can trigger inflammatory responses and activate the RhoA kinase pathway, leading to increased retinal vascular permeability and subsequent vision loss in diabetic patients. Therefore, developing potent antagonists that can modulate p75 NTR activity is essential for preventing diabetic-induced retinal damage. In this context, we will introduce a non-peptide p75 NTR modulator – (Rac)-LM11A-31.

**Definition**

(Rac)-LM11A-31 is an isomer of LM11A-31 dihydrochloride and serves as an orally active and potent proNGF antagonist that modulates the p75 neurotrophin receptor. According to the (Rac)-LM11A-31 description, this compound is utilized to inhibit the pro-apoptotic and pro-inflammatory signaling pathways associated with p75 NTR.

**In Vitro and In Vivo Studies**

The chemical properties of the compound are defined by the (Rac)-LM11A-31 formula C12H27Cl2N3O2, with a molecular weight of 316.27. Research focusing on (Rac)-LM11A-31 biological activity has demonstrated its efficacy in mitigating vascular complications. In vivo studies showed that the modulation of the p75 neurotrophin receptor using this compound prevents diabetes-induced retinal vascular permeability in mouse models. This protective effect is achieved through the inhibition of inflammation and the suppression of the RhoA kinase pathway, which are key drivers of retinal leakage in diabetic conditions. By blocking the interaction between proNGF and p75 NTR, (Rac)-LM11A-31 effectively preserves the integrity of the blood-retinal barrier. In conclusion, (Rac)-LM11A-31 is a potent proNGF antagonist that holds significant potential for the treatment of diabetic retinopathy.
